玉米根系分泌物中七种有机酸的LC-MS/MS快速定量分析方法研究
Study on rapid quantitative analysis method of seven organic acids in maize root exudates by LC-MS/MS
玉米根系分泌的有机酸在土壤养分活化、根际微环境调节及植物生长发育中具有重要作用。传统检测有机酸的方法(如HPLC、GC等)存在分离效率低、前处理复杂或灵敏度不足等问题,限制了有机酸的高效分析。本研究建立了基于LC-MS/MS的快速定量分析方法,采用HR-ODS色谱柱(150 mm×3.0 mm,3 μm),以0.2%甲酸水-乙腈(pH 6.5)为流动相(0.5 mL/min),优化了质谱参数,可在5 min内完成7种目标有机酸(L-苹果酸、柠檬酸、反式-乌头酸、DL-酒石酸、L-乳酸、丙二酸、丁二酸)的分离测定。方法检出限为0.08~29.12 μg/L,加标回收率84.10%~130.16%。应用于实际样品,成功检出4种有机酸(L-苹果酸4.169±0.21 μg/L、柠檬酸42.571±3.03 μg/L、DL-酒石酸16.274±1.12 μg/L、丙二酸797.391±15.32 μg/L)。该方法高效、灵敏、准确,为研究玉米根系分泌物组成及其生态功能提供了可靠手段。
Organic acids secreted by maize roots play a crucial role in soil nutrient activation,rhizosphere microenvironment regulation,and plant growth and development.Traditional methods for detecting organic acids (e.g.,HPLC,GC) suffer from limitations such as low separation efficiency,complex pretreatment,or insufficient sensitivity,which hinder efficient analysis.This study established a rapid quantitative analysis method based on LC-MS/MS,utilizing an HR-ODS chromatographic column (150 mm×3.0 mm,3 μm) and a mobile phase of 0.2% formic acid in water-acetonitrile (pH 6.5) at a flow rate of 0.5 mL/min.Mass spectrometry parameters were optimized to achieve the separation and quantification of seven target organic acids (L-malic acid,citric acid,trans-aconitic acid,DL-tartaric acid,L-lactic acid,malonic acid,and succinic acid) within 5 minutes.The method demonstrated a detection limit of 0.08-29.12 μg/L and a spike recovery rate of 84.10%-130.16%.When applied to real samples,four organic acids were successfully detected:L-malic acid (4.169±0.21 μg/L),citric acid (42.571±3.03 μg/L),DL-tartaric acid (16.274±1.12 μg/L),malonic acid (797.391±15.32 μg/L).This method is efficient,sensitive,and accurate,providing a reliable tool for studying the composition and ecological functions of maize root exudates.
