Multicolor fluorescent cell imaging technology for signal amplification and simultaneous detection of ATP and miRNA-21
基于氧化石墨烯(GO)的吸附承载能力与杂交链式反应(HCR)的无酶信号放大技术,提出一种多色荧光细胞成像方法,实现了对ATP和miRNA-21的同步检测与信号放大。该方法可在活细胞内对浓度差异达数个数量级的ATP和miRNA-21 2种生物标志物进行检测成像。实验结果表明,ATP和miRNA-21的检测限分别达到0.15 μmol/L和71 pmol/L。本方法有望为相关癌症诊断提供更精准的检测策略。
Based on the adsorption capacity of graphene oxide (GO) and the enzyme-free signal amplification technology of hybridization chain reaction (HCR),a multicolor fluorescence cell imaging method is proposed,achieving simultaneous detection and signal amplification of ATP and miRNA-21.This method enables the detection and imaging of two biomarkers,ATP and miRNA-21,with concentration differences spanning several orders of magnitude in living cells.Experimental results show that the detection limits for ATP and miRNA-21 reach 0.15 μmol/L and 71 pmol/L,respectively.This approach holds promise for providing a more precise detection strategy for related cancer diagnosis.
