HPLC法同时测定谷氨酸发酵尾液中8种有机酸的含量
Simultaneous determination of eight organic acids in glutamic acid fermentation tailings by HPLC method
建立了高效液相色谱法(HPLC)同时测定谷氨酸发酵尾液中8种有机酸含量的方法。对谷氨酸发酵尾液中有机酸含量进行测定,结果表明,样品经Copure SAX SPE固相萃取柱净化后,采用Agilent ZORBAX SB-C18(150 mm×4.6 mm,5 μm)进行分离,优化后的色谱条件为:光电二极管阵列检测器检测波长为210 nm,流动相为V(pH=2.7的磷酸二氢钾溶液)∶V(甲醇)=99∶1,流速0.7 mL/min,柱温35℃,进样量10 μL。在此优化条件下,8种有机酸在6 min内均能很好地分离和定量,各有机酸标准曲线相关系数R2>0.999 5,检出限为0.13~3.66 mg/L,定量限为0.44~12.20 mg/L,回收率为107.63%,相对标准偏差(RSD)均<5%。该方法线性关系良好,且简便快速、精密度高、准确度高,可用于谷氨酸发酵尾液中有机酸含量的快速测定。
A high performance liquid chromatographic (HPLC) method is established for the simultaneous determination of eight kinds of organic acids in glutamic acid fermentation tailings.Prior to be separated by an Agilent ZORBAX SB-C18 (150 mm×4.6 mm,5 μm),the samples are cleaned up by a Copure SAX SPE solid phase extraction column.The chromatographic conditions are optimized as follows:the detection wavelength of the photodiode array detector is 210 nm,the mobile phase is V(potassium dihydrogen phosphate solution at pH=2.7)∶V(methanol)=99∶1,the flow rate of mobile phase is 0.7 mL·min-1,the column temperature is 35℃,and the injection volume is 10 μL.Under these optimized conditions,the eight kinds of organic acids can be well separated and quantified within 6 min.The correlation coefficient of the standard curve for each acid R2>0.999 5.The limit of detection (LOD) ranges from 0.13 mg·L-1 to 3.66 mg·L-1,and the limit of quantification (LOQ) ranges from 0.44 mg·L-1 to 12.20 mg·L-1.The recoveries of eight organic acids are 107.63%.Relative standard deviations (RSD) are all less than 5%.This method exhibits good linearity,and is simple,rapid,precise and accurate for the rapid determination of organic acid content in glutamic acid fermentation tailings.
