摘要
通过对石油烃降解菌Paenarthrobacter ureafaciens Thp2-23进行16S rRNA基因测序分析,利用MEGA 7.0构建菌株系统发育树,并采用二苯碳酰二肼分光光度法检测Thp2-23在不同条件下对Cr (Ⅵ)的还原率;采用SPSS 25.0对各组还原率进行统计学描述及方差分析,探讨pH、温度、石油烃、吐温80和铜离子对菌株还原Cr (Ⅵ)效果的影响。结果表明,菌株Thp2-23在pH 8.0、40℃的最优还原条件下,12 h对50 mg/L Cr (Ⅵ)的还原率为95%,24 h对Cr (Ⅵ)的还原率达到100%。石油烃和吐温80可抑制Thp2-23对Cr (Ⅵ)的还原,铜离子可促进Thp2-23对Cr (Ⅵ)的还原。
Abstract
A 16S rRNA gene sequencing analysis is done on Paenarthrobacter ureafaciens Thp2-23, a petroleum hydrocarbon degradation strain.The strain phylogenetic tree is constructed by using MEGA 7.0.Diphenylcarbazide spectrophotometric method is employed to detect the reduction rate of Cr(Ⅵ) by Thp2-23 under different conditions.By means of SPSS 25.0 software, descriptive statistical analysis and variance analysis are performed on the reduction rates of each group.The influence of pH, temperature, petroleum hydrocarbons, Tween-80 and copper ion on the effectiveness of Cr(Ⅵ) reduction by the strain is explored.The results indicate that under the optimal reduction conditions including a pH of 8.0 and 40℃, the reduction rate of 50 mg·L-1 Cr(Ⅵ) by Thp2-23 strain reaches 95% within 12 h, and 100% within 24 h.Cr(Ⅵ) reduction may be inhibited by petroleum hydrocarbon and Tween 80, but promoted by copper ion.
