摘要

以三七(Panaxnotoginseng)植株为研究对象,分别对其根、茎、叶、花、整株中的三七素质量分数进行分析。通过微波-超声辅助提取工艺提取三七植株中三七素,用响应面优化对工艺进行优化,利用HPLC检测三七素质量分数。结果表明,三七花中的三七素质量分数最高,其次是叶,整株与根的质量分数接近,茎中质量分数最低。最佳提取工艺条件是:超声时间为 25 min、微波温度为80℃、料液比(g/mL)为1:31、超声功率为180 W、微波时间为10 min,该工艺条件下提取的三七素质量分数为30.77 mg/g,将溶液浓缩冻干得到三七素冻干粉质量分数达到46.59 mg/g。用MTT测定提取物对细胞活力的影响以及经LPS刺激后RAW264.7细胞NO释放量。结果显示,三七素提取物在10~200 μg/mL质量浓度范围内可显著抑制NO的释放,具有良好的抗炎活性。

Abstract

Taking Panax notoginseng plant as the research object,the contents of dencichine in root,stem,leaf,flower and plant are analyzed.Dencichine is extracted from Panax notoginseng plants through microwave-ultrasound assisted extraction.Response surface optimization method is used to optimize the extraction process,and the content of dencichine is determined by means of HPLC.The results show that the content of dencichine in the flowers of Panax notoginseng is the highest,followed by the leaves.The content of dencichine in the whole plant is close to that in the root,and the lowest in the stem.The optimum conditions are that the ultrasonic time is 25 min,the microwave temperature is 80℃,solid-liquid ratio is 1:31 g·mL-1,ultrasonic power is 180 W and the microwave time is 10 min.Under these conditions,the content of dencichine extracted is 30.77 mg·g-1,and the content of dencichine reaches 46.59 mg·g-1 after the solution is concentrated and lyophilized.The effect of extracts on cell viability is determined by means of MTT,and the released amount of NO from RAW264.7 cells after LPS stimulation is also detected.It is indicated that dencichine extracts can significantly inhibit the release of NO in the concentration range of 10-200 μg·mL-1,showing a good anti-inflammatory activity.
