摘要
从自然界中分离纯化出6株微藻,经过200 mL柱式光照反应器通气培养对微藻进行筛选,得到一株具有产油能力的优良藻株。实验结果表明,6株藻种油脂含量均在20%以上,生物量均超过1.2 g/L,其中SS6生长最快速,12 d进入稳定期,OD689达到12.270 3,细胞干重达1.337 g/L,油脂含量达到30.1%,油脂产率达到25.9 mg/(L·d)。经18S rDNA鉴定,SS6藻株为Monoraphidium komarkovae。对比研究多种保藏方法,表明SS6在-18℃、20%甘油保护下可保藏1年以上,复苏后其生长和产油能力均能保持保藏前的状态。
Abstract
Six strains of microalgae are isolated and purified from the nature.Among them,an excellent oil producing strain is obtained through screening and cultivating microalgae strains in a 200 mL column photobioreactor.Experimental results show that the total liquid contents and dry weights of six strains of microalgae all exceed 20% and 1.2 g·L-1,respectively.Among them,the strain SS6 grows the fastest and enters stable phase after 12 days in the photobioreactor.Its OD689,biomass,grease contents and grease productivity reach 12.270 3,1.337 g·L-1,30.1% and 25.9 mg/(L·d),respectively.Phylogenetic analysis by 18S rDNA identifies that SS6 is Monoraphidium komarkovae.By evaluating various preservation methods,it is found that SS6 can be stored for more than one year at -18℃ under the protection by glycerol (20% v/v).After recovery,its abilities in growth and oil production both can maintain the states before preservation.
